On my reconciliation all payments/deposit/cleared balance are correct - why then do I have a -£27,000 difference flagged up in right hand corner?

There are many reasons why reconciliation can show a difference when all payments are tricked.

Is this the 1st reconciliation?

Is the opening balance correct?

If it is not the first reconcile are there other transactions in the reconcile that do not fit in the date range that have auto ticked?

If that all seems okay go to the chart of accounts>account history on the account you're reconciling>click on the funnel symbol above the word date and change reconcile status to reconciled. Are there any transactions that show as already reconciled in that period?
